For the in‑vitro quantitative determination of Brazil Nut residues in food matrices such as bakery products, cereals, chocolate, cookies, crackers, flour, ice cream, milk, sausage, and yogurt. This test is designed for use by trained laboratory professionals in food testing, allergen monitoring, and quality control.
The Allergen‑Shield Brazil Nut ELISA (Cat. No. A2948/A2996) is a quantitative sandwich enzyme‑linked immunosorbent assay (ELISA) for the detection of Brazil Nut protein residues in food products labeled as Brazil Nut‑free. The assay detects trace contamination or deliberate inclusion of Brazil Nut protein and ensures compliance with international allergen labeling regulations (EU No. 1169/2011, USA, Canada, Australia, New Zealand).
Principle of the Test
The Allergen‑Shield Brazil Nut ELISA is a sandwich enzyme immunoassay using wells coated with specific antibodies against Brazil Nut proteins. Standards and samples are added to the wells, and any Brazil Nut proteins bind to the immobilized antibodies. After washing, a peroxidase‑conjugated secondary antibody is added, forming a complex. A chromogenic substrate (TMB) is introduced, producing a color reaction proportional to the concentration of Brazil Nut proteins present. The reaction is stopped with acid, and absorbance is measured at 450 nm.
Performance Characteristics
- Limit of Detection (LOD): 0.25 ppm Brazil Nut (0.009 ppm Brazil Nut protein)
- Limit of Quantification (LOQ): 1.0 ppm Brazil Nut (0.036 ppm Brazil Nut protein)
- Standard curve range: 1–25 ppm
- IC50: 7.0–14.0 ppm
- Recovery: 95.8% (CV = 6.9%)
- Intra‑assay CV at 5 ppm: 5.9% (n=16)
- Intra‑assay CV at 15 ppm: 6.4% (n=16)
- Strong reactivity to Brazil Nut (Bertholletia excelsa) with minimal reactivity to adzuki bean, navy bean, and rapeseed
- No cross‑reactivity up to 500,000 ppm with tree nuts, legumes, seeds, flours, meats, or spices
Storage and Stability
Store all reagents at 2–8 °C. Do not freeze. Protect colorless reagents (TMB substrate and standards) from light. Reseal unused strips with desiccant immediately after use. Expiry dates are printed on the labels. No quality guarantee applies after expiration or improper storage.
