Allergen-Shield Casein ELISA
QUANTITATIVE ELISA FOR CASEIN ALLERGENS

- Quantitative results in 30 minutes after sample preparation
- High specificity using monoclonal antibodies
- Uniform protocol across the Allergen‑Shield ELISA range
- Broad food matrix compatibility
- Surface swabbing kits available
- Excellent reproducibility and precision
For in‑vitro quantitative determination of casein residues in food matrices such as bakery products, beverages, bread mix, cake mix, non‑dairy chocolate, cooked biscuit, cookies, corn flour, dressings, ice cream, infant cereal, soy products, sausage, and wine. Intended for professional laboratory use in food testing and allergen control.
The Allergen‑Shield Casein ELISA is a quantitative sandwich enzyme‑linked immunosorbent assay (ELISA) for the detection of casein proteins in food products labeled as casein‑free. The assay identifies casein contamination or deliberate addition and supports compliance with international allergen labeling regulations (EU No. 1169/2011, USA, Canada, Australia, New Zealand).
Principle of the Test
The Allergen‑Shield Casein ELISA uses microtiter wells coated with specific monoclonal antibodies against casein proteins. Standards and prepared samples are added to the wells. If casein is present, it binds to the immobilized antibodies. After washing, a peroxidase‑conjugated detection antibody binds to the captured casein, forming a sandwich complex. A chromogenic substrate (TMB) is then added, producing a color proportional to the casein concentration. The reaction is stopped with acid, and the absorbance is measured at 450 nm.
Performance Characteristics
- Limit of Detection (LOD): 0.17 ppm Casein
- Limit of Quantification (LOQ): 0.5 ppm Casein
- Standard curve range: 0–30 ppm
- IC50: 4–14 ppm
- Recovery: 89.3% (CV = 8.1%)
- Intra‑assay CV at 8 ppm: 5.3% (n=16)
- Specific for bovine casein; cross‑reactivity with buffalo’s milk observed
- No cross‑reactivity with β‑Lactoglobulin or casein from goat/sheep milk
- No cross‑reactivity up to 500,000 ppm with tree nuts, legumes, seeds, flour, egg, fish, or crustacea
Storage and Stability
Store all reagents at 2–8 °C. Do not freeze. Protect colorless reagents (TMB substrate and standards) from light. Reseal unused strips with desiccant immediately after use. Expiry dates are printed on the labels. No quality guarantee applies after expiration or improper storage.
