For the in‑vitro quantitative determination of lupin residues in food matrices such as bakery items, cereals, chocolate, sauces, soups, and wine. The test is designed for use by professional laboratories for allergen control and quality assurance testing.
The Allergen‑Shield Lupin ELISA (Cat. No. A2648/A2696) is a quantitative sandwich enzyme‑linked immunosorbent assay (ELISA) designed for the detection of lupin residues in food products labeled as lupin‑free. The assay identifies trace levels of lupin proteins, ensuring compliance with international food allergen labeling regulations (EU No. 1169/2011, FDA, FSANZ, and Codex Alimentarius).
General Information
Lupin (Lupinus spp.) is a legume increasingly used as a protein‑rich ingredient in bakery, pasta, and meat substitute products. It contains allergenic conglutin proteins (α‑, β‑, γ‑, δ‑conglutins) capable of eliciting allergic reactions even at very low levels, including in individuals allergic to peanuts due to cross‑reactivity.
Principle of the Test
The Allergen‑Shield Lupin ELISA uses a sandwich enzyme immunoassay format. The microtiter wells are pre‑coated with antibodies specific to lupin proteins. Standards and extracted samples are added to the wells, and any lupin proteins present bind to these immobilized antibodies. After washing, a peroxidase‑conjugated detection antibody binds to the captured antigen, forming a sandwich complex. The addition of TMB substrate produces a color proportional to the lupin concentration, measured spectrophotometrically at 450 nm.
Performance Characteristics
- Limit of Detection (LOD): 0.25 ppm lupin (0.025 ppm lupin protein)
- Limit of Quantification (LOQ): 1.0 ppm lupin (0.1 ppm lupin protein)
- Standard curve range: 0–25 ppm
- IC50: 4–12 ppm
- Recovery: 92.4% (CV = 6.7%)
- Intra‑assay CV at 5 ppm: 5.8% (n=16)
- Intra‑assay CV at 15 ppm: 6.0% (n=16)
- Highly specific for lupin proteins
- No cross‑reactivity up to 1,000,000 ppm with tree nuts, legumes, seeds, flour, meats, or spices
Storage and Stability
Store kit and reagents at 2–8 °C. Do not freeze. Protect standards and TMB substrate from light. Reseal unused strips with desiccant immediately after use. Expiry dates are printed on labels.
