For the in‑vitro quantitative determination of sesame residues in food matrices such as bakery products, sauces, spreads, dressings, confectionery, and processed foods. This kit is intended for use by trained laboratory personnel in allergen testing and quality assurance laboratories.

 

The Allergen‑Shield Sesame ELISA is a quantitative sandwich enzyme‑linked immunosorbent assay (ELISA) designed for the detection of sesame residues in food products labeled as sesame‑free. The assay provides accurate quantification of sesame proteins to ensure compliance with international allergen labeling regulations (EU No. 1169/2011, FDA, FSANZ, and Codex Alimentarius).

General Information

Sesame (Sesamum indicum) is a widely used seed containing potent allergenic proteins such as Ses i 1, Ses i 2, and Ses i 3. These allergens are thermally stable and resistant to processing, capable of eliciting strong allergic reactions even at trace levels. Sesame is included among the priority allergens under major international food labeling standards, requiring sensitive analytical detection in both raw and processed food products.

Principle of the Test

The Allergen‑Shield Sesame ELISA is based on a sandwich enzyme immunoassay. Microtiter wells are coated with antibodies specific to sesame proteins. Standards and extracted samples are added to the wells, and sesame proteins, if present, bind to these immobilized antibodies. After washing, a peroxidase‑conjugated antibody binds to the captured antigens. The addition of TMB substrate produces a color reaction proportional to the sesame concentration. The reaction is terminated by the addition of acid, and absorbance is read at 450 nm.

 

 

Performance Characteristics

  • Limit of Detection (LOD): 0.25 ppm sesame (0.05 ppm sesame protein)
  • Limit of Quantification (LOQ): 1.0 ppm sesame (0.2 ppm sesame protein)
  • Standard curve range: 0–25 ppm
  • IC50: 7–14 ppm
  • Recovery: 90.3% (CV = 7.3%)
  • Intra‑assay CV at 5 ppm: 5.3% (n=16)
  • Intra‑assay CV at 15 ppm: 5.9% (n=16)
  • Highly specific for sesame proteins
  • Minimal reactivity to almond (0.02%), cashew (0.022%), hazelnut (0.012%), pecan (0.21%) and sunflower seed (0.025%).
  • At concentrations as high as 100,000 ppm, no cross-reactivity was observed with the other matrices including pine nut, walnut, peas, lentils, beans, pumpkin seeds and buckwheat.

 

 

Storage and Stability

Store kit and reagents at 2–8 °C. Do not freeze. Protect standards and TMB substrate from light. Reseal unused strips with desiccant immediately after use. Expiry dates are printed on labels.
