For the in‑vitro quantitative determination of total milk proteins (casein and β‑lactoglobulin) in food matrices such as chocolate, bakery products, dairy substitutes, beverages, sauces, and processed foods. For use by trained laboratory personnel in food allergen monitoring and quality control testing.

 

The Allergen‑Shield Total Milk ELISA (Cat. No. A1448/A1496) is a quantitative sandwich enzyme‑linked immunosorbent assay (ELISA) for the detection of milk protein residues, including caseins and β‑lactoglobulin, in food products labeled as milk‑free. The test provides rapid and accurate quantification of milk proteins, ensuring compliance with global food allergen labeling regulations (EU No. 1169/2011, FDA, FSANZ, and Codex Alimentarius).

General Information

Milk allergy is one of the most common food allergies, particularly in children. Cow’s milk contains approximately 3.2% protein, of which ~80% are caseins and ~10% are β‑lactoglobulin. These two protein groups are the primary allergens responsible for adverse immune responses. β‑lactoglobulin is more heat‑stable, making its detection crucial in processed foods. The Allergen‑Shield Total Milk ELISA detects both casein and β‑lactoglobulin for comprehensive allergen screening.

Principle of the Test

The Allergen‑Shield Total Milk ELISA is based on a sandwich enzyme immunoassay. Microtiter wells are coated with specific antibodies against milk proteins. Standards and extracted samples are added to the wells, allowing binding of milk proteins. After washing, an HRP‑conjugated antibody binds to the captured milk antigens. The addition of TMB substrate develops a blue color proportional to the milk concentration, which is measured at 450 nm after adding Stop Solution.

 

 

Performance Characteristics

  • Limit of Detection (LOD): 1 ppm milk protein (equivalent to 31.25 ppm milk or 3.125 ppm milk powder)
  • Limit of Quantification (LOQ): 2.5 ppm milk protein (equivalent to 78.1 ppm milk or 7.81 ppm milk powder)
  • Standard curve range: 0–67.5 ppm
  • IC50: 18–40 ppm
  • Recovery: 91.3% (CV = 6.4%)
  • Intra‑assay CV at 15 ppm: 5.1% (n=16)
  • Intra‑assay CV at 35 ppm: 6.6% (n=16)
  • Detects milk proteins from cow, goat, sheep, and buffalo milk
  • Minimal cross‑reactivity with non‑dairy proteins

 

 

Storage and Stability

Store kit and reagents at 2–8 °C. Do not freeze. Protect standards and TMB substrate from light. Reseal unused strips with desiccant immediately after use. Expiry dates are printed on labels.
